Uropein-based olive leaf extract enhances muscle mitochondrial bioenergetics response to moderate – but not maximal – intensity exercise in humans

Poor cardiorespiratory fitness is associated with several chronic diseases, many of which can be delayed or prevented by means of regular aerobic exercise. Two effective exercise routines are moderate-intensity continuous exercise (MICE) and sprint interval exercise (SIE). SIE has been shown to be equally or more effective than MICE despite a lower workload and shorter duration, which is partly due to differences in mitochondrial metabolism leading to pyruvate dehydrogenase (PDH) activation. The authors hypothesised that administering natural polyphenol oleuropein from olive leaf extracts (OLEs) may activate PDH and potentiate mitochondrial adaptations.

They carried out a crossover, double-blind study where healthy participants completed MICE or SIE routines with either placebo or an OLE dietary supplement. Muscle biopsies were collected before, immediately after, and 24 hours after exercise; samples from the placebo group were labelled with TMT-6plex. Mascot Server 2.8 was used for peptide and protein identification, followed by downstream analysis in Scaffold Q+ 5.

Of 1497 proteins identified, MICE was shown to have modified the levels of 174 proteins immediately post exercise and 4 proteins after 24 hours. SIE modified the levels of 15 proteins immediately post exercise and none after 24 hours. The most upregulated pathways immediately after both MICE and SIE were immune and inflammatory responses, while mitochondrial-related pathways were somewhat down regulated – consistent with previously reported decrease in mitochondrial respiratory capacity after high-intensity exercise. The effect of using OLE was then investigated using RNAseq. This revealed effects on the mitochondrial and inflammatory response after MICE and SIE, but an increase in PDH activity only after MICE.
